HEALTH-2009-2.2.1-1 Synaptopathies: genesis, mechanisms and therapy. FP7- HEALTH-2009-single stage
Our project aims at utilizing oxide nanoparticles as free radical scavengers in neurodegenerative processes focusing our action on retinal diseases, which can lead to blindness, Alzheimer's disease and brain ischemic insults. The rationale is to develop a new, simple and inexpensive therapeutic tool to be used alone or in conjunction with other more traditional therapies. The advantage of cerium oxide nano particles treatment is to be a pure antioxidant tools with no collateral effects.
1) Nanoparticles preparation seems to be critical. In our models only one type was effective in counteracting photoreceptor loss.
2) We have to verify whether nanoparticles might interfere with central nervous system functions and development (to test toxicity is not enough).We are a network of three research units with specific expertise to cope with this challenge.
Two Research units in our university:
Department of Physic and NANO_CAT srl (spin off company of the university) : Nanoparticles preparation: The CeO2 nanoparticles were prepared by the precipitation method from cerium nitrate and ammonia and subsequent annealing at various temperatures up to 800°C. The as-prepared precursor powders and calcined powders were studied by various characterization tools.Department of Science and Biomedical Technology : Nanoparticles testing on animal models of neurodegenerarative diseases. Electrophysiological recording can be performed both “in vivo” and “in vitro” from retina and different cortical areas, including the entorhinal cortex, either during acute application of nanoparticles in the bath solution or in chronic application.

EXPERTISE REQUIRED:
- Under our coordination the requested partner(s) shoud be able to develop methods of nanoparticles functionalization with appropriate chemicals not poisonous for the human body to be used as carriers to deliver the particles in the different region of CNS. It would be of interest to have nano into eye drops to reach the neuroretina or dispensed intranasally as nasal spray for transporting to the brain areas by means of the olfactory neural pathway.
